| Résumé : | Artificial inoculation of conifer seedlings by selected beneficial ectomycorrhizal fungi is now widely used in forest nurseries. However, very little information is currently available on the dissemination and persistence of these inoculated strains. For example, the inoculated fungus could be replaced by indigenous strains and the beneficial effect of the controlled inoculation be lost. Monitoring the persistence and dissemination of introduced symbionts is difficult due to the limitations of the identification methods. Recently, molecular techniques, based on PCR amplification, have been developed to identify ectomycorrhizal fungi on a single root tip. Here, we have used the polymorphism of the PCR-amplified ribosomal DNA intergenic spacer (IGS) to identify Laccaria bicolor S238 on inoculated Spruce and Douglas fir in a forest nursery and in a plantation. Roots of Douglas fir seedlings and Norway spruce cuttings were mostly (80-90 %) associated with L. bicolor S238 after two years in the nursery. Two years after outplanting in a forest site, L. bicolor S238 was still present on the spruce root system, but represented only 18 % of the mycorrhizal symbionts indicating a high competition by the indigenous species. |
